About This Product

■Achieve high productivity

High Productivity Harvest (HPH) is a new application developed by Repligen. Fed-batch bioreactors can be harvested in completely sterile conditions for processing in collection columns. It also eliminates both centrifugation and depth filtration.

■Improvement of productivity

Increased throughput by taking the same number of days to collect the same titer

■Process improvement

・Suppress process-related impurities to achieve healthier culture ・Reduced number of unit operations and raw materials ・Collection of materials that can be immediately processed downstream

■Improvement of VCD and survival rate by HPH

・Compared to fed-batch, HPH fed-batch achieves 4 times the viable cell density ・Survival rate maintained close to 100%

■Improvement of product titer with HPH

・Twice the productivity compared to conventional fed-batch ・Collection in the same number of days (or less) as fed batch

■Mechanism

It consists of three main steps: starting the fed-batch process, installing the cell retention device for High Productivity Harvest, and drawing down the bioreactor. HPH start dates are flexible and vary by process. Harvesting can be completed in the same time frame as traditional fed-batch, or bioreactor runs can be extended to further increase yields depending on process and equipment constraints. ・Step A: Start the process Start the fed batch process as usual. ・Step B: CRD attachment and High Productivity Harvest Once peak cell density is achieved, attach the cell retention device (CRD) and pump in fresh medium at a low flow rate. Product collection begins slowly as fresh medium begins to be injected. ・Step C: Drawdown On the final day of the run, finish adding fresh medium and begin drawing down the bioreactor capacity. ・Step D: End of process HPH fed-batch can be completed in the same time frame as traditional fed-batch or can extend production time to further increase yields.
