Cytotoxicity test (NK cell activity, LAK cell activity, CTL, ADCC, etc.)
This is a device that uses fluorescent staining to perform tests that are generally performed using radioactive isotopes (51Cr). Target cells are labeled with a fluorescent dye (Calcein AM) instead of 51Cr, and after reaction with effector cells, the fluorescence intensity of the remaining target cells is used as an indicator of cytotoxicity, and shows a high correlation with the 51Cr release method.
■Features
・Video recording of cytotoxicity is possible.
- By incorporating a digital camera system, it is now possible to observe and record damage to target cells over time.
・Use time-lapse (optional) to create videos and images of cell damage in specific wells.
・Using a high-resolution epifluorescence microscope photometer, cytotoxicity can be measured with high sensitivity even in minute amounts of samples.
・It has many advantages as it does not use RI.
・No special facilities or troublesome considerations such as half-life, storage, usage amount, environment, etc. are required.
・High sensitivity requires 1/5 to 1/10 fewer cells.
・Measurement time after reaction is short.
- Significantly reduces running costs.
・Kinetic measurement based on time is possible.
・The degree of injury can be visually observed and confirmed under a microscope, and images and videos can be recorded.
■Applications
LAK/NK activity, CTL, ADCC, cytotoxicity test in cell-mediated immunity/humoral immunity.
