About This Product

■About the measurement target/sample DNA

・Can detect UV-induced damage to biological DNA (cyclobutane-type pyrimidine dimers (CPD) and (6-4)-type photoproducts (6-4PP). Sample DNA must be extracted from cells. . ・This product can be used for DNA of all living things. Ultraviolet rays are classified into three types depending on their wavelength. They are called UV-C (100-280nm), UV-B (280-320nm), and UV-A (320-400nm) in descending order of wavelength (stronger energy). UV-C (100-280 nm) and UV-B (280-320 nm) are directly absorbed by DNA within cells, exciting the conjugated double bonds in the bases. This results in the frequent formation of cyclobutane-type pyrimidine dimers (CPD) and (6-4 type) photoproducts (6-4PP). UV-A (320-400nm) is also directly absorbed by DNA and forms CPDs, but at a much lower rate. However, 6-4PP does not form. Solar ultraviolet rays that reach the earth's surface become mixed waves of UV-B and UV-A because the ozone layer absorbs UV of 290 nm or less, including UV-C. Among CPD and 6-4PP formed by UV-B, 6-4PP is efficiently converted to DewarPP (Dewar-type photoproduct) by UV-A irradiation.

■Notes

・When using an ELISA kit, it is necessary to extract DNA from cells. You can also use commercially available DNA extraction kits. The antibodies themselves can also be used for immunohistological staining. ・The purity of DNA used as a sample is recommended to be 2.0≧A260/A280≧1.8. (A260 260nm absorbance measurement value, A280 280nm absorbance measurement value) ・DNA samples do not need to be fragmented with restriction enzymes, etc. ・The amount of DNA damage (CPD/6-4PP) cannot be measured using an ELISA kit. ・Cannot be stored after solution preparation. Please use the OPD Substrate Solution prepared immediately before use.

■How to denaturalize DNA (separate it into single strands)

・Dispense the diluted DNA sample into tubes (with cap locks). ・Heat for 10 minutes in a heat block set to 100℃. - Immediately cool on ice (0℃) for 15 minutes.
