■A BREAKTHROUGH TECHNOLOGY

Phenotypic microarrays (PM) are the third technology in the genetic domain for research and drug discovery (Figure 1), following DNA microarrays and proteomics technologies. DNA microarray and proteomics technologies have made it possible to analyze the level of thousands of genes and proteins at once. Phenotypic microarrays allow quantitative measurements of thousands of cell expression systems at once. DNA microarray and proteomics technologies have made it possible to detect genes and proteins that regulate patterns of change that have important correlations with disease states. However, there was no guarantee that these changes would be significant in cells. Phenotypic microarrays are a complementary technology that provides the information needed at the cellular level. Phenotypic microarrays provide comprehensive cellular profiles for gene function identification, drug target evaluation, optimization and toxicity studies. After genetic changes or exposure to a lead drug, the intracellular response of the changes can be directly assessed. Currently, phenotypic systems can be used for microorganisms and fungi. We are also developing plates for mammalian cells.

■HOW PM TECHNOLOGY WORKS

Phenotypic microarrays are a breakthrough technology. This is a cell assay system that integrates equipment and software for cell HTS. The technology and testing process is shown in Figure 2 below. BioLog preconfigures a wide range of expression system tests in array sets. Each well of the array contains test media for a different expression system. By simply placing pre-cultured standard cell suspensions into each well of the microarray, you can test various types of expression systems at once. Microarrays are typically incubated for 24 hours.
