■Summary

Myocardial cell activity stain-free measuring device CIOS8. Just place the culture plate on this device. It is possible to measure the activity of cardiomyocytes in culture without staining and non-invasively. The measurement results are displayed in real time as a waveform, and the pulsation period and contraction time are automatically detected with the touch of a button. Built-in temperature control and plate movement mechanism. It is small and lightweight, making it ideal for personal use.

■Main features

・Automatic detection of heart muscle cell pulsation and contraction time without using fluorescent dyes or electrodes ・By using the built-in fluorescence filter, simple membrane potential measurement using membrane potential sensitive dyes such as Di-4-ANEPPS is also possible.

■Measurement method and principle

The measurement principle of this device is very simple. A light-emitting diode placed on the underside of the sample illuminates the sample brightly and obliquely. It is well known to researchers in the field of optical measurement in biophysics that cellular activity is accompanied by changes in light scattering, and it is known that contraction of the heart muscle produces a relatively large signal. The reflected light component is received by a photodiode, and only the changing component is amplified by a high-power amplifier and extracted. This signal is sent to a digital processing circuit via an ADC, which magnifies the slightest optical change by a factor of several thousand to tens of thousands of times, and performs numerical processing to identify it as myocardial activity.
