■Summary

3D Cell Explorer-fluo, a holographic and fluorescence microscope for cell observation, is an innovative microscope that combines 3D refractive index imaging and 2D fluorescence imaging using holography and tomography technology. This is a revolutionary microscope that allows you to quickly and easily visualize the internal structure of cells and tissues, as well as the distribution of organelles. By overlaying a refractive index image with a fluorescent image of various cell organelles and cell membranes, it is possible to calibrate digital staining so that the image is the same as the fluorescent label. This digital staining calibration allows the 2D fluorescence image to be expanded into a 3D image. Additionally, this calibration process allows correlation between fluorescent staining and digital staining, making it possible to handle unstained samples. The constructed 3D image can be scaled, rotated, and displayed in cross section, allowing you to observe the inside of the cell from various viewpoints.
