Adipose stem cells can be isolated from a small amount of human adipose umbilical cord tissue in a short time, easily, and with minimal damage to cells.

■About stem cell separation substrate

When tissues such as human fat, umbilical cord, and bone marrow are cultured on a stem cell isolation substrate (hereinafter referred to as the substrate), stem cells (hMSCs) outgrow along the substrate. It takes advantage of the properties of tissues, which are difficult to adhere to flat structures such as flasks, and easily trapped in fibrous structures. By trapping and culturing tissues on a three-dimensional structure base material made of PE-PP (polyethylene-polypropylene) core-sheath nonwoven fabric coated with hydroxyapatite, stem cells that produce abundant extracellular matrix can be isolated. Masu. In addition, since stem cells proliferate faster than fibroblasts and vascular endothelial cells, it is possible to isolate highly pure stem cells by limiting the culture period.

■Features

・Easy to operate in a short time: Just cut the tissue into small pieces, place it on the stem cell separation substrate, and culture it. ・Can be isolated if there is a small amount of tissue: Stem cells can be isolated from human fat about the size of a grain of rice, umbilical cord tissue (approx. 20 mg), and human liposuction fragments (approx. 0.5 mL) ・Highly pure stem cells: When cells collected from the separation substrate were cultured in adipocyte differentiation induction medium, it was confirmed that more than 80% of the cells differentiated into adipocytes. ・Less damage to cells: Since no collagenase is used, damage to cells can be minimized. ・Confirm differentiation potential and retention of cell surface markers

■How to use (simple version)

- Treat the base material to be hydrophilic and place it in a 6-well plate. ・After washing the adipose tissue or lipoaspirate with physiological saline (or PBS), place it on the base material. For liposuction, place the base material on the attached cell strainer and then place the liposuction ・Add 5 mL of medium to each well so that the separation substrate is submerged in the medium. ・Culture in a 37℃ incubator for 10 to 14 days ・When adipose stem cells occupy about 10-20% of the separation substrate, collect the cells. *Please perform the work in a sterile environment such as a safety cabinet. Tweezers, dishes, culture equipment, etc. are required.
