■About stem cell separation substrate

・A nonwoven fabric that can separate adipose stem cells from a small amount of human adipose umbilical cord tissue. ・Hydroxyapatite is applied to a nonwoven fabric with a core-sheath structure of PE-PP, and by placing human adipose tissue on this base material and culturing it, human adipose stem cells that produce abundant extracellular matrix can be isolated. ■Primary culture

・Hydrophilize the base material and place it in a 6-well plate ・After washing the adipose tissue or lipoaspirate with physiological saline, place it on the base material. In the case of liposuction, place the base material on the attached cell strainer and then place the liposuction ・Add 5 mL of medium to each well so that the separation substrate is submerged in the medium. ・Culture in a 37℃ incubator for 10 to 14 days ・Collect cells when adipose stem cells occupy approximately 10-20% of the separation substrate

■Cell collection and passage

・Transfer the substrate with outgrown cells to a 50 mL conical tube and wash twice with approximately 10 mL of physiological saline or PBS. If there are multiple substrates carrying the same specimen, it is possible to combine them into one 50mL conical tube for each two substrates. ・Add approximately 1 mL of cell detachment solution per substrate to the conical tube containing the substrate so that the substrate is submerged. ・Cells will peel off if you incubate at room temperature for about 5 to 10 minutes, so add about 20 to 30 mL of physiological saline or PBS and mix thoroughly by inverting. ・Remove the substrate and centrifuge at 1,200 rpm for 3 minutes at room temperature. ・Add the prepared growth medium for adipose stem cells to the obtained cell pellet and count the number of cells by pipetting.

■Cell morphology/proliferative ability

We confirmed the proliferation of hASCs when human adipose tissue was cultured in BSCM-SP1 adipose stem cell isolation medium using stem cell isolation substrate. - From day 5 to 6 of culture, the presence of hASCs outgrowing along the fibers of the substrate can be confirmed. Difficult to find when there are few cells ・After 9 to 10 days of culture, it can be clearly seen that hASCs are outgrowing along the substrate. ・After 10 to 14 days of culture, it proliferates throughout the substrate. Collect ASC from the substrate when hASC accounts for about 10-20% of the entire substrate.
