A new type of microscope incubator from BLAST that can be controlled to provide low oxygen levels has been released.
■Three features
1.No nitrogen gas is required and the oxygen concentration can be reduced to a nominal 0.3%, allowing the set concentration to be precisely maintained.
2.It is ultra-small and can be mounted on a microscope, making it easy to observe live cells in hypoxia culture.
3. Oxygen concentration can be changed minutely, making it easy to perform assays under a wide variety of hypoxic conditions.
■Gas reserve system that generates low oxygen gas
The control device has a reserve chamber (gas reservoir) for regulating gas. A deoxidizing agent is placed in the chamber to generate low-oxygen gas, which is then circulated within the chamber to create a low-oxygen environment. Oxygen concentration is maintained at a desired value by mixing with the atmosphere.
It is much simpler and has lower running costs than conventional low oxygen control using nitrogen gas. The gas reservoir is located at the rear of the control unit. Oxygen absorber lasts about 2 weeks. CO2 cartridges last for 2 months.
■Cantilever opening/closing structure
The culture chamber can be easily opened and closed with a single action using a cantilever. The sealed structure with a glass heater on the top and non-evaporated glass on the bottom ensures visibility while maintaining a high gas barrier and preventing atmospheric oxygen from entering.
■Oil immersion compatible version
A commercially available cover glass with a thickness of approximately 0.2 mm is exposed from the bottom, making it possible to perform high-magnification observations such as oil immersion and confocal observation even in a sealed state. The vertical and horizontal dimensions of the chamber are the same as the well plate, so it fits into a general plate holder without modification.

By installing it in various imaging equipment, it becomes possible to analyze Hypoxia in a continuous environment.

■Stop fitting
When the air supply tube is removed, it closes and oxygen leakage can be reduced to about 0.3% in 10 minutes (depending on the environment and culture conditions). Used for short trips.
■Branch fitting
Low oxygen gas is sent to two chambers in parallel, and one control device controls the gas concentration in the two chambers.
