■About albumin free
Animal-free refers to a medium that does not contain human or animal-derived materials, and albumin-free refers to an animal-free + recombinant medium that does not contain albumin. If the culture medium contains raw materials derived from humans or animals, there is a risk of virus infection, so this is a highly safe culture medium that eliminates these risks.
In addition to human and animal-derived materials, we use an albumin-free medium to minimize the use of plant and microbial-derived materials.
■How to use
・After opening, please add antibiotics directly to the culture medium bottle before use.
・Add serum each time you culture (we recommend not adding serum directly to the culture medium bottle)
■Cell morphology
We confirmed the proliferation of hASCs (human adipose-derived stem cells) when human adipose tissue was cultured in BSCM-SP1 adipose stem cell isolation medium (supplemented with 1% human serum) using stem cell isolation substrates.
- From day 5 to 6 of culture, the presence of hASCs outgrowing along the fibers of the substrate can be confirmed. Difficult to find when there are few cells
・After 9 to 10 days of culture, it can be clearly seen that hASCs are outgrowing along the substrate.
■Proliferation performance comparison
hASCs and hWJ-MSCs were cultured in BSCM PL1 medium (1%FBS) and basal medium (10%FBS) up to 8 passages, and the number of cells at each passage was measured. Showed excellent cell growth in PL1 medium
■Cell surface markers
As a result of confirming the expression of cell surface markers of hASCs and hWJ-MSCs after 6 passages, we found that surface markers (CD105,44,29,73,90) characteristic of mesenchymal stem cells (MSCs) retained their expression. surface markers characteristic of blood cells (CD45,14,31) were not expressed.
■Differentiation ability
After 6 passages, hASCs and hWJ-MSCs were induced to differentiate into adipocytes, osteoblasts, and chondrocytes, and the results showed that they differentiated into each cell type.
