About This Product

■Summary

NanoTemper's two unique technologies (microscale thermophoresis and Spectral Shift) accurately and quickly capture the interaction between two molecules and provide it as Kd. Since it does not have a flow path, regular maintenance is not required, and Kd can be determined with a very simple operation. Monolith is a standard type that measures with a capillary, and Dianthus is a high-throughput type that measures with a plate.

■Microscale thermophoresis

Heat is applied to a sample in a solution, and changes in fluorescence intensity due to thermal equilibrium movement of the target molecule and changes in the fluorescence intensity of the molecules themselves are captured, and changes in fluorescence intensity over time are measured. The degree of this change in fluorescence intensity changes depending on whether the ligand is bound or not. Measure the change in fluorescence intensity for each ligand concentration, draw a sigmoid curve based on the difference in the changes in fluorescence intensity, and calculate Kd.

■Spectral Shift

When a ligand binds to a target, the fluorescence spectrum changes. In this measurement technique, the target is labeled with a fluorescent label, a fixed amount is mixed with a series of dilutions of the ligand, excited at 590 nm, and the binding state is detected as a shift in the fluorescence spectrum. To detect this movement, fluorescence is detected at two wavelengths, 650 nm and 670 nm, and KD is calculated by plotting the ratio of the fluorescence intensities on the vertical axis and the ligand concentration on the horizontal axis. Since ratios are used in the KD calculation process, the calculation formula is (1). Therefore, noise can be canceled out by calculation, improving the S/N ratio. This makes it possible to detect even subtle shifts with great precision.

■Quick measurement
