The retention mechanism in reversed-phase chromatography is the opposite of that in normal phase (such as silica gel). Adsorbents in normal phase chromatography are polar and adsorb through polar interactions. Therefore, more polar samples are more strongly adsorbed and have lower Rf values ​​than less polar samples. It is possible to increase the Rf value by increasing the polarity of the developing solvent (mobile phase). In reversed-phase chromatography, the adsorbent is a relatively non-polar material and is adsorbed through non-polar interactions. Therefore, the less polar samples are strongly adsorbed and have lower Rf values ​​than the more polar samples. Generally, it is possible to increase the Rf value by lowering the polarity of the developing solvent (mobile phase). Miles Scientific's reverse-phase UNIPLATES™ uses hydrocarbon-impregnated silica gel. The hydrocarbons are absorbed onto the surface of the silica gel and are not chemically bonded to the silica particles. Impregnated reversed-phase UNIPLATES™ offer many advantages over chemically bonded PR18 TLC plates. Most importantly, it is compatible with aqueous solvents and is compatible with most C18 HPLC packing materials. Reversed-phase UNPLATES™ allows the use of 100% water as both the mobile phase and spotting solvent, making it very useful when working with polar samples such as proteins and peptides. Solvents containing water, buffers, and polar organic solvents (methanol, acetonitrile, THF, etc.) can be used, but using ethyl acetate or less polar or non-polar solvents will cause the hydrocarbons to escape from the silica gel. I can't use it because it's gone.

If resistance to organic solvents is required, use the reversed-phase Unibond™ HPTLC, a high-performance chemically bonded plate.
