Normal human monocytes are found in the peripheral circulation. Normal human monocytes play an important role in host defense during circulating monocytes and differentiation into tissue macrophages, and their strong antigen-presenting capacity allows them to differentiate into dendritic cells in vitro and in vivo. . Monocytes are isolated from the peripheral blood of healthy, screened donors. Peripheral blood is collected from the donor using a blood component separation device. The mononuclear cell-rich cell product is further processed by density gradient centrifugation to remove red blood cells and neutrophils. CD14 monocytes are then isolated using the immunomagnetic bead method. ■Recommended medium A serum-containing medium is recommended to preserve the monocyte phenotype (medium containing 10% FBS (fetal bovine serum) is generally recommended). It is also possible to add M-CSF (10 ng/ml). If producing osteoclasts, soluble RANK ligand (30 ng/ml) and M-CSF (30 ng/ml) must be added to the serum-containing medium. If producing dendritic cells, use a serum-free medium such as Lymphocyte Proliferation Basal Medium-3 supplemented with GM-CSF (50 ng/ml) and IL-4 (50 ng/ml) LGM-3™ Must be. ■Applications Cytotoxicity test
