Human natural killer (NK) cells are lymphocytes of the immune system that are essential for host defense and immune regulation. Since it is part of innate immunity, sensitization is not required for activation. NK cells play important roles in viral infections, autoimmunity, pregnancy, cancer, bone marrow transplantation, and, according to recent studies, adaptive immunity. NK cells are usually characterized by their response to the surface marker CD56 and non-reactivity to CD3. Poietics™ human NK cells are isolated by positive or negative selection using immunomagnetic bead technology. Purity is >90% analyzed using flow cytometry based on CD56 antigen response. CD16 expression has also been reported. Negative selection of NK cells recovers significantly more CD16+ cells (typically >80%) than positive selection of CD16+ cells, which is more variable. Cells isolated from positive selection are called natural killer T cells. ■Applications Cytotoxicity test
