■Human stromal cells When bone marrow cells are cultured under specific conditions, a stromal cell layer of adherent cells develops over several weeks. The stromal cell layer is composed of diverse cell types such as fibroblasts, mesenchymal stem cells, adipocytes, endothelial cells, and macrophages. The stromal cell layer, which acts as a feeder layer for hematopoietic progenitor cells, allows continued proliferation and differentiation of progenitor cells under the same culture conditions without the addition of exogenous cytokines. Long-term culture systems allow in vitro models of the bone marrow microenvironment, such as cell-cell interactions, adhesion molecules and cytokine secretion, to be studied. These and other factors tightly regulate blood cell production, progenitor cell contribution and differentiation, and stem cell regeneration.
